Culture media
Lowenstein-Jensen Medium. An egg-potato base solid media containing malachite green (an inhibitory agent). The use of L-J media is excellent for the recovery of M. tuberculosis from sterile-site specimens as well as decontaminated-digested sputum specimens.

Lowenstein-Jensen Pyruvate Medium. Pyruvate is an intermediary organic acid metabolite in glycolysis and the first of the Embden Myerhoff pathway that can pass readily into or out of the cell. Thus, its addition to culture medium provides both an energy source and a carbon skeleton for anabolic processes. Its addition may help in maintaining certain and may help when when the serum concentration is reduced in the medium. Pyruvate in LJ medium encourages the growth of M. bovis.

Acid Egg Medium. The least expensive of all TB-specific media available and supports good growth of tubercle bacilli. Contamination during preparation is limited because it is inspissated after being placed in bottles. In addition, the malachite green added to the media suppresses the growth of non-mycobacterial organisms

Pyruvic Acid Egg Medium.
- Supplied as pre-poured slopes in 50 x 7ml universals (CB39/0260), 25 x 7ml heavy universals (CB39/0260) and 25 x 5ml heavy bijous (CB39/0260)
Kirchner Liquid Medium. A liquid culture medium which is made selective by the addition of antibiotics and has been shown to be sufficient in the recovery of mycobacteria from non-sputum samples.

Stains
Ziehl-Neelsen. Staining according to the Ziehl-Neelsen reference method for acid-fast bacilli.

- Individual stain for ZN available: Carbolic Methylene Blue as 125ml, 1000ml and 2500ml (CB28/310100)
Kit BK-Colour. The Ziehl-Armand stain is a fast variation of the traditional ZN stain and gives the advantage of simultaneously decolourising all bacteria except mycobacteria that have taken up the carbol fuchsin stain.
- Supplied as Kit BK-Colour: 1 kit = 1 x 100ml of Ziehl carbol fuchsin and 1 x 100ml of Armand solution, providing sufficient reagents for 125 to 200 slides (CB28/364600).

Cold AFB stain. This cold AFB stain offers the advantage of not having to heat slides and producing toxic carbolic fumes. The stain is very quick and suitable on all types of substrates (sputa, cultures, etc). Sensitivty is equal to that of traditional ZN staining. Mycobacteria appear pink against the blue background of the slide.

Kit Quick-TB
Fluorescent Auramine stain. Fluorescent staining according to the Degommier is a fast technique that helps detect AFBs. Stained AFBs appear fluoresent yellow-green against a red background of the slide. Fluorescent staining allows fast screening and easy reading of slides. It is essential to confirm fluorescent-positive AFB by staining the same slide with a Ziehl technique.
- Supplied as Kit Fluo-Color: 1 kit = 1 x 250ml of Carbolic auramine, 1 x 250ml Degommier discolouring solution, 1 x 125ml Thiazine red concentrated solution and 1 x 125ml Carbolic solution (CB28/361460).

Phage-based Assay for TB Detection
A novel application of phage amplification technology, which allows the rapid detection of TB within 24 hours of sample preparation. The technique involves infecting mycobacterial cells present in the sample with a specific mycobacteriophage then treating the sample with a virucide to destroy any extracellular mycobacteria. Intracellular phage undergo replication and are released from host cells into a lawn of susceptible cells within an agar plate. The phage undergo further replication to produce plaques on the surface of the agar, indicating the presence of TB.

FastPlaque TB™ Kit
Phage-based Assay for Detection of Rifampicin-Resistant TB
Extension of the FastPlaque TB™ Kit (above) allows early detection of multi-drug resistant tuberculosis. A smear-positive sputum specimen is processed in a similar manner to the FastPlaque TB™ assay but preincubated overnight with and without rifampicin. Viable TB bacilli in the original specimen are reflected as plaques in the lawn of sensor cells. Rifampicin-resistant TB strains survive drug treatment and produce plaques, whilst sensitive strains do not.
